The design and synthesis of a new class of magnetic nanoparticles (MNPs) based on a perfluoropolyether (PFPE) surface coating, having citrate groups, is reported herein. A systematic approach was carried out to optimize the bulk and nanosized PFPE coating of MNPs, to be used as contrast agents, to improve their biocompatibility, and to generate stable colloidal dispersions. This was done by varying the preparation parameters and we observed that the resulting nanosized particles exhibited an average hydrodynamic diameter of about 175 nm, excellent colloidal stability, a saturation magnetization of about 43 emu g(-1), and excellent dispersion in water. The PFPE surface coating not only provided excellent colloidal stability to the MNPs but also created a more hydrophilic and biocompatible surface (as determined by blood coagulation and cytotoxicity studies) than bare MNPs. PFPE MNPs showed high transverse relaxivity values (r(2)) at a low iron concentration, which can be conveniently exploited in magnetic resonance imaging (MRI). Furthermore, the PFPE-coated MNPs do not aggregate after 30 days, indicating excellent colloidal stability for in vivo studies.
